Yes, of course. Installing the .obb: 1) Download the original game from PlayStore. 2) Go to /Android/obb and look for the game's obb folder. 3) Move it to somewhere you can easily remember. 4) Uninstall the game and then, place the .obb back to /Android/obb. 5) Then, install the modded APK.

There are no conditions to activate the mod. How are you testing it? All Enemies/Monsters stats should be 0/1 in quests.
