I have iPad Mini 5 on the version 16.4.1, I want to make OTA update to 16.5 but there's an error in the settings (Error message) because sometime what I was on iOS 14.4.1, I made am unc0ver jailbreak and enabled a function in the settings of unc0ver named "Disable Updates", so now I don't know how to disable it. Can someone help me?

If you're no longer jailbroken and cannot update due to you may having the "Disable Update" unc0ver feature lingering on your device, then you may have to update via iTunes on your computer.

Alternatively, a full restore would resolve this issue and remove any leftover jailbreak files since you have not restored rootFS before updating your iOS version.
