The new jailbreak doesn't have Mobilesubstrate installed. the reason why its not working with it. wait for the update i think its already out. Check the forum for 2.1.0 Jailbreak TaiG. Also with the previous jailbroken device reboot it. check if the game assist is installed. should help. by the way which source did you get it from?

You don't need to toggle anything. once installed make sure no in app tweaks are installed in cydia. reboot device. make sure you have the correct version installed. Should work. Working for me as of now.

Yes it does. The game assist is uninstalled after the maintainance. Re install it from game assists and it works just fine.
