I noticed that Sideloadly provides misleading information if you try to use a paid developer account with 2FA and "Use Anisette authentication" disabled. It says "App-specific passwords are not supported". However, when Anisette authentication is not used, ONLY app-specific passwords work for accounts with 2FA.

I don’t think this is the case. 

2FA should be supported regardless of free or paid Apple ID.

And paid Apple ID supports normal & app-specific passwords if I recall correctly.

We will investigate your issue to verify everything is working fine though!
